1962 948cc.
This is a strange one.
I recently altered the fuel mixture to rectify some slightly rough running.
It was then that I noticed the very top nut, the ssecuring one, had come loose and had turned with the adjustment nut.
I retightened the securing nut, checked the adjustment nut was 2 full turns down, but since that moment, she refuses to start.
is it possible that I have disturbed the jet centre or something similar?
Battery new, spark at plugs.
I am really puzzled.

Must have disturbed the jet. Re-centre it and check the needle is at the right height and free and comes all the way down and hits the bridge at the bottom. Bingo.

You have probably jammed the needle by putting the jet off centre. As above - recentre and check the piston rises and falls easily.
